Interleukin-17 (IL17/IL17A) is a pro-inflammatory cytokine produced by T helper 17 (Th17) cells. Because of its crucial roles in immune regulation, IL17 is now a target for an array of therapeutic monoclonal antibodies and inhibitors to treat various inflammatory diseases and cancer.
IL17 belongs to a distinct cytokine family with five additional members: IL17B, IL17C, IL17D, IL17E/IL25, and IL17F. IL17 shares the most sequence homology with IL17F. IL17 and IL17F form homodimers and heterodimers (IL17A & IL17F) to signal through the receptor complex composed of the subunits of IL17RA and IL17RC. Both IL17 and IL17F are major contributors in recruiting neutrophils to sites of inflammation. The IL17 and IL17F signaling can induce the production of cytokines (IL-1β, TNFα, G-CSF, IL-6, etc.), chemokines (MCP-1/CCL2, CXCL1, CXCL2, CCL20, etc.), inflammatory effectors, and antimicrobial proteins. The IL17RA subunit is used by at least two other ligands, IL17C and IL17E. Other members of the IL17 Receptor family include IL17RB, IL17RC, IL17RD, and IL17RE.
