Discover Rabbit Monoclonal Antibodies Against Challenging Targets

Traditionally, rabbits have been recognized for their ability to produce antibodies with higher specificity and affinity compared to mice and rats. However, the lack of a myeloma cell line for rabbits as a fusion cell strain made it challenging to use the hybridoma method to produce monoclonal antibodies. FUJIFILM has developed a new platform which overcomes these issues, combining the strengths of the DNA immunization method with their proprietary single B-cell screening technique to produce high-performance, high-affinity monoclonal antibodies against soluble antigens or transmembrane antigens. The antibodies produced using this platform are not only high-performing but also capable of recognizing complex three-dimensional structures and challenging targets such as multipass transmembrane proteins.

Advantages of Rabbit Monoclonal Antibodies

Rabbit monoclonal antibodies offer several benefits over traditional mouse monoclonal antibodies, including higher specificity and affinity for target antigens. The rabbit immune system generates a diverse antibody repertoire, enabling the recognition of a broader range of epitopes, which is particularly advantageous for detecting small molecules, sugar chains, and epitopes that may not be immunogenic in mice.

DNA Immunization Method

The DNA immunization method has a high success rate in generating antibodies against membrane proteins and complex conformational epitopes, which are often challenging to target using conventional peptide or protein antigens due to their inability to mimic native protein structures. By using DNA constructs, we can bypass many of the issues associated with traditional peptide/protein immunization techniques. Simply by altering the DNA sequence, we can generate mutants, deletion variants, or even express proteins from different species. This flexibility is particularly useful when working with complex antigens.

Single B-Cell Screening at FUJIFILM

FUJIFILM’s proprietary Single B-Cell Screening method enables the acquisition of high-affinity antibodies by directly selecting individual B cells that produce these antibodies against the target antigen, utilizing a unique application of EV technology. Single B-Cell Screening also results in more positive clones, providing a broader pool of high-quality antibodies to choose from.

Service Workflow

Our service follows a structured, milestone-based workflow:

  1. Feasibility Study: Analysis of the target protein’s structure and characteristics to develop a tailored antibody production strategy.
  2. Antigen Expression Analysis: Cloning the target cDNA into our in-house expression vector, followed by transfection into mammalian cells to verify target protein expression.
  3. Immunization and Serum Testing: Introducing the expression vector into rabbits and assessing antibody production against the target protein using flow cytometry with antiserum.
  4. B-Cell Cloning and Screening: Harvesting spleen cells from immunized rabbits, isolating antigen-specific B cells using our proprietary single B-cell cloning technology, and amplifying antibody genes from each B cell.
  5. Monoclonal Antibody Production: Expressing recombinant antibodies and evaluating their antigen-binding capabilities through flow cytometry or ELISA.
  6. Deliverables: Providing the antibody gene sequence and comprehensive reports detailing the development process and antibody characteristics.
