SolisFAST® product range – fast solutions, quality results
There is a saying: “Slow and steady wins the race”, but what if you could go fast and have an even bigger win? This is what the SolisFAST® range will give you with its fast reaction speed, inhibitor tolerance, and room temperature stability.
How fast is SolisFAST®?
SolisFAST® products contain oligo-hot-start DNA polymerase which is activated with as short as 20-30 seconds of denaturation treatment. There is no non-specific amplification during the reaction setup since the reversibly blocked and specially modified DNA polymerase won’t work until heated to 95 °C in the PCR device. The SolisFAST® enzyme itself has a 3-4 times faster extension rate (2-4 kb/ min) than a regular Taq DNA polymerase. As an extreme example, some clients have used a 1 s extension step for 1000+ bp targets and received results without any problems.
To save more of your time it’s also possible to choose between singleplex and multiplex options. With fast cycling endpoint PCR you can go up to 18-plex which will give you results in 39 minutes. With fast probe-based qPCR you can go up to 5-plex and get your results in 30-40 minutes, but if you would really like to test the limits then it’s possible to do the reaction even faster.
What is SolisFAST®’s contamination prevention strategy?
Contamination can ruin your day or even week. Data is not valid, a lot of cleaning must be done and supplies are wasted. Precious time and even samples can be lost. A combination of proactive methods and good laboratory practice helps us prevent contamination and perform accurate and sensitive analyses. As a part of the prevention strategy, carryover contamination from previous amplification of the same target sequence can be degraded by Uracil-N-Glycosylase (UNG). To make this method work, an amplicon has to contain Uracil residues. This is accomplished by including dUTPs in all amplification reactions. Since natural DNA sequences do not contain Uracil, then only the Uracil-containing DNA contaminants are degraded during the UNG treatment step and cannot be amplified in further analyses. It is advisable to use polymerases that can incorporate 100% of dUTP.
SolisFAST® DNA polymerase very effectively incorporates dUTPs compared to regular Taq DNA polymerases. You can also be well assured that your RNA sample is not degraded during the UNG treatment! UNG is not able to degrade naturally Uracil-containing RNA sequences – other types of enzymes are needed for this. During UNG treatment step contaminants are degraded and cannot be amplified in further analyses.
Is SolisFAST® really inhibitor tolerant?
Solis BioDyne’s SolisFAST® DNA Polymerase is an in silico designed enzyme with fast extension rates and a programmed tolerance to inhibitory substances. Combining that with an optimized buffer, the SolisFAST® qPCR Mixes enable robust qPCR performance and accurate target detection under demanding conditions. To better understand SolisFAST® qPCR range’s inhibitor tolerance capabilities, Solis BioDyne has put together a white paper. It includes results from different tests, shows real-life application possibilities, and feedback about the products. You can read it HERE.
What are the other benefits of SolisFAST® product range?
Thanks to the Stability TAG technology, all the products are room temperature stable and can be shipped without ice, making packages, shipping fees, and your carbon footprint smaller.
