KRAS Nucleotide Exchange Assay

Their TR-FRET-based Nucleotide Exchange Assay enables precise measurement of GTP binding to KRAS in the presence of SOS1, a key guanine nucleotide exchange factor (GEF). The assay utilizes a Terbium-labeled anti-Tag2 antibody (HTRF donor) to detect fluorescence-labeled GTP binding to KRAS. Upon excitation at 340 nm, energy transfer occurs, producing a measurable fluorescence signal at 665 nm. This assay provides a quantitative method to evaluate nucleotide exchange activity, with inhibitors reducing the HTRF signal.

KRAS-cRAF Binding Assay

The TR-FRET-based KRAS-cRAF Binding Assay is designed to detect interactions between activated KRAS (loaded with GppNHp) and cRAF. The assay employs a Terbium-labeled anti-Tag2 antibody (HTRF donor) for KRAS and a fluorescence-labeled anti-Tag1 antibody (HTRF acceptor) for cRAF. When KRAS binds to cRAF, fluorescence resonance energy transfer (FRET) occurs, enabling quantitative measurement of the interaction.
